Dave, Is there any chance that you moved the serial adapter to another USB port, or that you plugged in other USB devices since this last worked?
I've had this problem before and it was a result of my serial adapter getting assigned to a different device in the OS (such as, hypothetically /dev/tty1 instead of /dev/tty0). That usually results from plugging in to a different usb port.
