As a general rule you should shut down <all> programs (including Office Notifications) before any backups. Copying an open file is an invitation to disaster. FileMaker Pro databases, if copied while open, will, when the copy is opened, tell you that the file was not closed properly and go into a file checking routine, thus circumventing possible problems, but most applications are not that careful and will just barf on the corrupted file.
