The OS doesn't really care where the files are on disk, it just reads and writes them. However, after a while the disk contents can have files scattered throughout the disk, forcing new files to be split into multiple pieces. Most UNIX systems use a disk format that doesn't *really* require defragging, but OSX uses HFS+, which can benefit from periodic defragging (a couple of times a year or so). -- Glenn L.
