You should be able to upgrade to Exchange5.5 Enterprise with no changes to
your domain. It is pretty straightforward (choose re-install, and then
reapply the service pack you were at and all patches). 

Other than that, the offline defrag may work, if there is enough free space
in your .edb (I'd check your most recent App Event ID 1221 for the
public/private databases to see how much free space was reported by the last
online defrag).

You will want to check on licensing issues if you care about such things,
since enterprise is a more expensive license (and 5.5 isn't sold anymore).
