For storage on the server, start the Service Manager, go down to the private store and look at the mailbox table. It will show you the Total items per user. Add them up and do the math.
Now for SMTP, that is a bit more work. The one way I know is to enable SMTP logging for a day or three, then review the logs. We have logging enabled always and review them for anomalies every week. Also if it is included in EVERY message you have just increased the storage needed for every message in the message store. It does start to add up after a while.
