Personally I would strongly recommend considering a service like
MessageLabs (www.messagelabs.com) or Exchange Hosted Services Filtering
(http://www.microsoft.com/exchange/services/filtering.mspx). 

The two biggest advantages are keeping the spam traffic off of your
Internet connection and having someone else manage the system to ensure
maximum filtering while minimizing false positives.

That said, if you're set on running it yourself Iron Port has a good
reputation, but I haven't worked with their products or any other
anti-spam appliances.

Hello, we are currently evaluating the Iron Port entry level C series
anti-spam product. Anyone out there used it before, anyone like it, hate
it? So far it looks pretty good. It's a 1U server and it's supposed to
handle up to 1 million emails per hour. It's also supposed to do 90% of
the spam checking in memory so it won't be writing much to disk. We are
currently using GFI mail essentials/mail security and we absolutely hate
it, bad performance, bad anti-spam engine, horrible support.

We are looking for an ISP grade anti-spam solution as with a two letter
domain we get hit pretty hard.
