Large brushes are cruel & unusual punishment - they really load up the CPU and produce 
slow screen updates, even on a fast machine with lots of RAM. It's the amount of data 
that needs processing quickly when using a large brush. If PS is hitting the disk 
instead 
of working in memory, it's going to be ever so slow. You need more RAM and/or faster 
CPU 
and/or faster graphics card and/or PS memory % settings need experimenting with. 
Probably 
the whole lot, or use a smaller brush and stay solvent;).
