You might try setting things up as per

Though you will want to edit /sw/bin/xinitrc.sh as a superuser and change

x_prefix="/usr/X11R6"

to

x_prefix="/opt/X11"

And possibly also use

#!/bin/sh
. /opt/X11/lib/X11/xinit/xinitrc.d/98-user.sh

as your /sw/etc/xinitrc-last-hook--though I'm not completely sure that's
the location of the file on Xquartz on 10.6.
