Absolutely sure. It happened several times during a series of hardware
problems with one of our mail servers.  I was proxying two mail domains but
not a third (a personal domain I host work).  Mine was always OK, the others
bounced whenever the RoBoX didn't see the servers.  This was up to at least
3.6.x.- but this is on an elderly RoBoX-R unit that has some other quirks.

I do a/v and a/s on the mail servers and have our ISP perform inbound and
outbound a/v and a/s (as well as store & forward))on the pipe.

My tests show a 4xx error being issued which should generally cause the 
sending mail server to queue the message.

I did test through a relay server though (the relay server queued the 
message just fine when the firewall couldn't reach the backend mail
server).

Overall the AS/AV works quite well in our experience.
