For make.conf vs make.defaults yes but I am talking about package.use
I figured any USE setting in package.use for a specific pkg would override
any general USE setting in make.conf or make.defaults.
For package.use in /etc/portage this is true but not for 
package.use located in the profile.

No, I can not subtract USE flags in profile/package.use if the same
flag is present in make.conf
