The area polygon must be the only one in the view\group. If not create a new group. Edit the group containing the shape file. Select the contour of the area, copy it and paste it in the new group.
Then, Map Tools --> CAD Tools --> Calculate Area of Closed Polygons and select the group containing only the limits of the area. You will have surface in square meters, if the coordinate units are in meters. Dividing the area by 100m, the line spacing, will give the total length of the survey in meters. Divide by 1000 to get it in km. The total length includes only the flight line inside the limit.
