(Maybe it is a feature I have turned off?) But regardless, you can type "is:unread" in the search box at the top of your screen, which will show all unread messages that are in "All Mail". It's not a button, you have to type it. But you can use the Quick Links Gmail Lab and make it a link that you can click. I am confused what the "unread mail" question has to do with sending replies, because both the message you are replying to, and the reply itself, would be considered read.
