Click-Drag-Release is a de facto way of defining an "area of interest". I guess you can try to train the users of a particular application to do differently. Click to initialize tells the system your clear intent and provide a precise definition. If your first drag did not fit exactly what you want, you can get a better rectangle in second attempt easily while holding the shift key. It's hard to do so using the no-click approach because you have no idea when the rectangle starts drawing, or you have to release/press the shift key all the time. Just my 2cents. Maybe a poll will help.
