I looked through a few books (Liber Usualis and Graduale Romanum 1974) and
it seems from a cursory look that there is always at least the space of a
punctum between the clef and the first note. When the vowel is not the first
letter of a word, there the text often goes underneath the clef at the
beginning of a line, but always with there being a bit of room between the
clef and the note.

I'd be happy to do a more careful comparison if this might be helpful, but
the space of a punctum or a punctum and a half might be a good rule of
thumb. Does anyone else have thoughts on this?
