FYI: think of Growl sort of like an e-mail client.  Even if you have the e-mail 
client downloaded and installed, if no one sends you e-mail you don't have 
anything to read in the client... just a big, empty inbox.  Growl provides a 
centralized system for nice, configurable notifications from applications, but 
if you don't have any applications set up to generate those notifications, then 
there's not a lot for it to do.

Trillian should work out-of-box with Growl.  You can check whether or not 
Trillian's even /seeing/ Growl by going to Trillian -> Preferences, 
Notifications.  If all the notification options are greyed out, Growl isn't 
being picked up.  Make sure you /are/ running build 1.2.2, the latest build!  
If that's not enough to get you working on Trillian, talk to me off-list and 
I'll try to get you sorted (I'm the developer).  

Unfortunately, Skype uses an outdated version of the framework, so you'll need 
to use GrowlVersionDetective -- available from http://growl.info/downloads -- 
to upgrade the framework.  That'll get it up and running until Skype releases a 
new version with a less cobwebby old version of the framework.

For Mail.app, there's a third-party plugin called GrowlMail which generates 
Growl notifications when new mail arrives; you can find GrowlMail at 
http://code.google.com/p/growlmail/downloads/list as a free download.
