Ah, if GrowlMail works, you don't need to use growlnotify to test or anything else. (Looks like you got GrowlMail working while I was writing my long mail.) If you had Trillian running already when you installed Growl, it may for some reason have not properly registered once Growl was up and running; if that's the case, quitting Trillian and restarting it would work (and then I'll add a bug report to my own tracker to look into why that would happen). If that /doesn't/ work, I can try to help you off-list in a bit. (I need to go get dinner shortly, so I'll be away from the keys beginning soon.)
