Windows key X to bring up the mobility center, then the key m to unmute.
This works on my notebook running Vista.

Perhaps hitting the "v" once you have entered the systray will bring you
to the Volume item in the list. There can't be too many items that begin
with "v" in the systray.

Not all system trays are the same. My speakers were at the top of my
list.
