At work: One volume. Even with our technical user base, I'd rather just avoid problems with users not putting files in the correct locations.
At home: I generally used 20-25GB for XP, and use 50-55GB for Vista. It just makes reinstalls easier and quicker, since you have less data to move off before nuking...even though I may only do them once every year or two. I also do automated boot volume backups every other night, making a reasonably-sized boot volume nice. Not to mention that keeping your OS files closer to the outside of the spindle (first part of a disk to fill up) by creating a smallish boot partition effectively short-strokes the drive, which can result in faster boot times.
