I assume you've tried rebooting the server and that didn't help.
And take a look at your imail log file. Try to send a message
from the web with a known to and from address and then check the
log file. If there is nothing there at all then imail1.exe
didn't fire. If there is something there that will be more
information.
One good possibility is that permissions have changed for iusr
and it no longer can execute imail1.exe or it might not be able
to write a message file used by imail1.exe. So either way check
permissions. If you have logging on your nt server then check
your event log otherwise turn it on and then check.
It might possibly be a firewall or proxy issue too but I think
the permissions is the most likely possibility right now.
