Have you tried 'installing' Leo to a hard drive using 'python setup.py install' or the windows installer and then just copying the Leo folder under site_packages to the equivalent place under portable python and running launchleo.py.
I have Leo running under portable python - so it does work - at least on windows. There isn't anything special that needs to be done.
