Bonjour,
Finally, the change is not persistant, so I've made a file called 
molette.sh like that :

#!/bin/sh
xsetwacom set cursor RelWUp "button 5"
xsetwacom set cursor RelWDn "button 4"

And do a "chmod 777 molette.sh" and put it in /usr/bin/

Then in the gnome menu system->pref->sessions, I have added this command 
line "sh /usr/bin/molette.sh" to the list of prog that must be launched 
at the beginning of the session.

It works, and it's enough waiting for the bug (?) solution.
