How about %-based widths (fluid layout) and em/%-based font sizes. No
dependency on screen width what so ever.

If you want  a truly elastic design, ie the design fills the browser window by adjusting the font-size (and the whole website including images are in ems) see my little experiment on my blog http://nickcowie.com/2006/the-fluid-elastic-reboot/
Warning it is rather orange and still needs some work, especially if you default font size is larger than 16px.

It is a little radical but you could use http://nickcowie.com/xtras/fontsize4.js to form the basis of a script that changes font size based on browser window width, so users get one version of the website at 800px wide screens and another on 1000px wide screens.

There is a difference between screen resolution and browser width, though in my experience most users still running 800x600 screens have their browser window maximised. And that was a decent sample of "ordinary" users, 200 staff in a government office.
