Another advantage to Firewire drives is Target Disk Mode, which allows you to migrate your files to a new computer easily if you're not able to use Migration Assistant for some reason (I was not, and Target Disk Mode saved me).

USB will work fine for Super Duper. I use them all the time. Firewire will just work faster.

Also, older PowerPC Macs can’t  boot off USB drives.
