I did a quick test in desktop Mail.app (Mavericks). When you do a "Subject contains" search, it does find the string in the middle of the subject line, but only if it is a prefix of a word.

I think Spotlight has this behavior as well.

Yup. I entered "afari", got very few results, changed it to "safari" and the list contained many more results, including this very discussion thread. :)
