Well, I just checked the same shortcut, once assigned to "Go to Favorite Box 
[bla]" and once assigned to "Flag [b]".

When assigned to "Go to Favorite Box [bla]" it *systematically refuses to work 
after a Mail relaunch and I have to manually activate it by accessing the menu 
item first. I've tried Fullscreen and not fullscreen, automatically hide menu 
and not, and all configurations produce the same result.

When assigned to "Flag [b]", it *systematically* works without requiring 
anything.
