You can split up an iTunes library across multiple disks, as long as those 
disks are consistently accessible under the drive letters or volume names with 
which they were identified at the time you added their contents to your iTunes 
library.  The trick is in iTunes Advanced Preferences; uncheck the "Copy files 
to iTunes Media folder when adding to library” checkbox.  Now, adding files to 
your library from any disk you care to will create a reference to the external 
files on those disks, without building the library in your primary iTunes 
location.

Be aware that this scheme creates for you the obligation of making certain that 
the files continue to be available when you need them, for example when 
synchronising.  Moreover, and perhaps more significantly, you lose the ability 
to organise files automatically, which means that any change to metadata that 
you make to files inside iTunes will not be reflected in the file system, but 
only in the file tags and iTunes database.  This last reason is why I could 
never consider doing this.  It would be far better to make a virtual file 
system that spanned multiple disks, or delegate storage to a larger drive 
and/or NAS that will be represented as a single file system.
