On my Yosemite iMac Startup Disk is already unlocked.  It’s controlled from 
Security & Privacy, General tab, Advanced button, and the funny checkbox that 
changes its name.  Make sure it’s unchecked so it says something like “Some 
System Preferences with lock icons will be unlocked”, or some such thing.

You can also use the bless command, but on El Capitan you have to turn SIP off 
in order to use it.  “man bless” for details (requires command-line foo).
