Do you want to use Cygwin, MSYS, or MSYS2? MSYS provides a POSIX-like environment for running configure scripts and POSIX tools. The user experience resembles a GNU system - configure, make, make install. Althought there are, of course, special things (like giving it the right prefix, understanding FS layout, etc). The only version of MSYS is provided by mingw.org, use their mingw-get installer to install it. It's quite old though, and has a very limited set of [old] applications. MSYS2 is the new hotness. Behaves like MSYS, but is based on Cygwin, and is thus very up to date. I know of two MSYS2 vendors - alexey (the current MSYS2 maintainer) and me. Cygwin is...well...Cygwin. It can only cross-compile (if you want to build binaries that are not tied to Cygwin), which may or may not be what you want. MSYS and MSYS2 can be used with mingw-w64 (the steps you need to take to integrate mingw-w64 with MSYS or MSYS2 depend on their vendors; i know how to integrate mingw-w64 with MSYS, MSYS2(alexey's) and MSYS2(mine)).
