Correct. Many japanese software of the "good times" (until 1992) uses this
method: disk stations, pink sox, peach up, xak's...

When booting, MSX BIOS reads sector 0 of the disk in drive A in a fixed
memory location, and executes it. Normally, code for loading MSXDOS(2).SYS
and COMMAND(2).COM is placed here, so DOS starts. But you can also put in
this sector code for read other sectors via BIOS routine #0144, so no FAT
nor directory structure is needed.

This is a problem for installing such software in hard disk: you must
convert these disks to disk image files, and if you are lucky (most times
this means: if you have MegaSCSI) you can execute it via special programs.
