Follow-up Comment #1, bug #15136 (project mypaint):
I would prefer layers modes rather than brush modes, simply because on layer
with blending mode u can use all brushes, otherwise we would be forced to
change each brush blending mode individually.
Also we can easily modify layer opacity or delete it if we don't like
changes. With brush blending modes it's impossible (unless we do undo lots of
times :/ and loose part of painting we wanted to keep).
Also cool tricks can be made with layer like overlay:
if we duplicate current layer and set it blending mode to overlay we would
have image with increased contrast, and opacity of this layer would increase
or decrease this contrast. More examples here:

Of course we don't have possibility to duplicate layers, but maybe in
future...
