Open drain screws on each bowl and make sure fuel comes out each one. Fresh fuel and a remote tank. If using regular tank, pull vacuum on the petcock vacuum port to prime carbs.
The choke on these aren't really a choke but a fuel enrich system. Those circuits can get clogged up. A thin guitar E string is my weapon of choice when checking these. When in doubt, break em back down and clean, sucks but better to spend the time redoing and making sure all is clean. Could also be spark on one or more. Either lay plugs on engine to ground them or use a jumper wire to engine. It's a little spark. Hard to see in sun light. I'd lean more towards fuel. Also make sure carbs are not over filling and flooding engine and washing down cylinders, this takes the compression out as the fuel removed the oil between rings and cylinder walls. The bike will crank faster than normal if this happens.
