Do you mean your customer user or the agents who reply to tickets?

Customer can create tickets via the webinterface (customer.pl). Also it
is possible to fetch customer messages via pop3 or pipe messages
directly into OTRS via the PostMaster.pl module. How this works depends
on your setup and on the operating system you use.
Agents only can process tickets via the web interface. They can not
answer, forward, close, e.g. tickets by email.
