yes. you lose the benefit of initial conversion in 16-bit/channel mode and the added freedom of making a variety of adjustments during conversion. going from 12-bit/channel to 8 and then to 16 has lost lots of shadow detail. going directly from 12 to 16 retains it. this matters if you spend some time manipulating your images before finalizing them.
