Yes, you can use it that way.  Telnetting to port 25 allows you to
send raw SMTP commands to diagnose the SMTP server (whatever it might
be).  Heck, you can telnet to port 80 and send raw HTTP/GET requests
to any web server even...

Which is why the telnet client remains such a useful diagnostic tool
even if the telnet remote login service is considered harmful...
