Here's one idea Have you been using the lens with the additional screw thread for the second cable release used for mirror up operation? Sometime this thread remains protruding out from the lens barrel even AFTER removing the cable release, and so the lens won't fire via the usual shutter release button. Reconnect the mirror up cable release, and then undo it again: A gentle push as you do this ensures the protruding bit goes back flush with the lens barrel.
