Open the TDB file via "File->Load Map". Next you will be asked for a basemap. 
That will be the img file that covers the complete area of the map. The 
detailed tiles will be found by the information in the TDB file.

Using contour lines to calculate tour profiles is a very poor approach. The 
commonly used gradient algorithm are poor for tricky mountain areas. But as you 
have created the contour lines you must have a source with the elevation 
information. If that source is in a format understood by GDAL you can attach 
the data to the map by doing a right click on the map in the left hand list and 
select "Add DEM...".

In the route tab (the one with the upward arrow and the point) is a tab with a 
"?". That explains how to calculate routes vie online services. But keep in 
mind your Garmin will not care about what QLGT calculates. It will calculate 
it's own route with a result of it's own.

The resulting overlay line can be converted to a track or a route. And vice 
versa. In fact if you want to tweak the position of points in a track or a 
route you have to convert it into a distance line.
