I can't vouch for all-day rides I don't do, but I agree with the others that a 
good bike with the saddle and bars in the right places is the key to painless 
riding. In general I've found that padding made me less comfortable on the 
bicycle; even Riv's formerly-padded Summer Gloves made me feel like I was 
trying to push my hand through this big THING in my palm to get to the bars. 
I've been much more comfortable since I switched over completely to Brooks 
saddles, wool underwear with Riv shorts, and unpadded gloves. I used to think 
Spencer-at-Riv was crazy to ride that beausaged Rivendell Road without bar 
tape, but I'm starting to see his point.
