I know about the non-supported. But in my experience it works fine.
I did one (using yum, not up2date) to go from redhat 9 to RHEL 3, without a problem.

I would not do it for a really essential server, but for a 'test machine' it is good enough. Our company plan is to go from RHEL 3 to 5 next year for all systems, and early experience with RHEL 5 could be an advantage. Just for one test server, after all. In the worst case we can always do a fresh install on it when the time comes.
