Hi Ralf,
did you change anything in your configuration?
Did you try to restart the x_server? 
'File cannot be locked' occures i.e. if an NFS mount is used without the
'nolock' mount option. Did you checked that?
Even if it sounds very unlikely, did you checked setuid root of lserver and
dbmsrv?
Another possibility is that another replication manager is already running,
that still locks the file. (Use ps -ef | grep lserver to check that).
