[Histonet] p16 Antibody

We have been using Ventana's p16 antibody and detection kit on an IHC stainer 
that is not Ventana's stainer.  We have been told that these reagents are now 
obsolete, but that they can be purchased in dispensers which are customized for 
Ventana's own stainer.  Can I simply just dispense their reagents into my own 
vials for my stainer?  I'm wondering if anyone else is dealing with this same 
issue.

Are there other companies that sell the p16 antibody?  The last time I checked 
(which was years ago), Ventana was the only one selling this.

We monitor the concentration and pH of our recycled formalin weekly, but since 
it has been consistently within range I may start monitoring monthly.

[Histonet] Silver disposal

I'm wondering how others dispose of their silver waste from a GMS stain?  We 
have always collected and disposed of it separately from our other chemical 
waste.  We have a 55 gallon drum where we dispose of formalin, alcohol and 
xylene, and I was told by several people that we could just add the silver into 
the 55 gallon drum since it was such a small percentage of silver.

We use 10 ml of a 5% silver solution and then dilute that to 50 ml, so there is 
a very small percentage of silver in the solution.

[Histonet] Special stains and hoods

I would like to know how many people perform (manual) routine special stains 
under a hood.  We do basic specials - Giemsa, AB/PAS, GMS, Trichrome, Muci - 
but we do them on the counter, not under a hood.  We have been told the 
staining needs to be done under a hood, but I have never done that in my entire 
career.

We have three Thermo 355S's.  They are nice because you can use them in the 
automated mode or manual mode.  All of our HT's use the manual mode, so now we 
just buy the manual version.

We use the Diff Quik in place of the Giemsa stain for H. Pylori on gastric 
bx's.  I believe you can use the Diff Quik for FNA's, especially if it is just 
for a preliminary diagnosis and you will be staining with H or a cytology 
stain later.  We get our Diff Quik from Medical Chemical Corporation.

for fine needle aspirates. Recently, when we tried to reorder a Diff Quik stain 
kit from our usual suppliers, we have found it to be on backorder. This has 
caused us to begin to consider the Giemsa stain as an alternative to the Diff 
Quik. Does anyone have any experience with using both stains and how they 
compare to each other? Would the Giemsa be a suitable alternative to replace 
the Diff Quik?

When you say "labels" are  you talking about paper labels or the print on the 
slides from the Slidemate??

I will assume that you mean the print, and yes, I have had the problem in the 
past where the print came off during staining and/or you could literally wipe 
the print off with a kimwipe or your finger.  In our case, it was the slides - 
even though it was the same slide we had been using for years.  This was 
happening with a certain lot number.  Is this happening with just one lot 
number?  Maybe you can try and get a different lot number or try a totally 
different slide (different manufacturer) to narrow down the issue.

Hello all,

I am hoping someone out there can help. We have been having issues with some of 
the labels coming off during staining and after. It seems to be random so I 
can't tell if there is a problem with the slides or the Slidemate AS. There are 
days that it will be half the slides but other days where it will be none. 
Sometimes the labels peel off and float away during staining and other times 
labels rub off when handling but only after staining.


We are using Globe Scientific Slides, and have been for over 2 years with no 
issues until December.

I have changed out the ribbon on the slidemate and the problem is still 
sporadic.

Oven time drying time does not seem to be a factor.

We have not made any reagent changes for the staining process to explain the 
issue either.


Has anyone else seen anything like this? Any slide suggestions to try out?
