CHARM Therapeutics is establishing a state-of-the-art R facility in Cambridge, UK. We are currently seeking a motivated Biochemist to join the Structural Sciences group at CHARM Therapeutics. The position is within a lab-based team, working on high throughput crystallographic and CryoEM structural targets to fully enable and drive multiple SBDD / Med Chem campaigns. The successful candidate will join a multidisciplinary, highly collaborative discovery team to invent novel medicines and drive them towards the clinic. Role Requirements: The successful candidate will be expected to meet the following criteria: A degree in biochemistry / molecular biology, or a related discipline, is required. Experience in a biotech/pharma setting may be advantageous. Molecular biology / cloning / protein engineering skills and knowledgeable in expression construct design, and downstream purification strategies. Demonstrable experience in the expression and purification of target proteins from recombinant systems (both bacterial and eukaryotic advantageous) utilising various affinity systems. Experience in the use of FPLC systems and different approaches to purify recombinant target proteins to homogeneity. Knowledgeable in the application of protein biochemistry to aspects of drug discovery programs including structural biology and biophysical analyses Excellent organisational / interpersonal skills with the ability to work as part of a team. Role We are currently seeking an experienced tissue culture scientist to join the structural sciences group at CHARM Therapeutics at the level of Associate Principal Scientist, establishing, managing and maintaining our tissue culture laboratories utilising multiple expression systems to drive high throughput crystallographic structure determination across multiple SBDD campaigns. Requirements The successful candidate will be expected to meet the following criteria: Minimum of a strong honours degree plus a post graduate qualification up to, and including PhD level qualification in biochemistry / molecular biology, or a related discipline, is required. Additional previous experience in an industrial / biotech setting would be preferred. Extensive expertise and knowledge of recombinant protein expression systems; sterile cell culture techniques, protein biochemistry and downstream purification strategies and upstream molecular biology skills in terms of PCR, cloning, bacmid generation and sequencing. Experience in both transient transfection methods and baculovirus mediated expression protocols. Demonstrable track record of setting up, using and maintaining multiple and routine large scale expression systems - including bacterial, insect cell and mammalian adherent and suspension cell cultures. Experience in performing extensive quality control on protein expression runs., i.e. Western blot analysis, SDS-PAGE etc. Excellent organisational skills in terms of ordering tissue culture stocks and planning for expression run logistics. Excellent interpersonal skills with the ability to work as part of a team. Proven track record in presenting results (both written and verbal) to internal and external audiences. Ability to solve and refine crystallographic structures including expertise in using CCP4, Global Phasing and Phenix crystallographic software suites. Deep understanding of SM / NCE ligand chemical restraint definition and subsequent ligand co-structure refinement. Demonstrated proficiency in shell scripting and experience in developing pipelines to handle large scale diffraction data processing. Experience with PanDDA approaches for data processing would be beneficial. Proven track record in presenting results (both written and verbal) to internal and external audiences. Extensive practical experience and expertise in the expression and purification of multiple target proteins from recombinant systems (both bacterial and eukaryotic) utilising e.g. cutting edge 3rd Gen affinity systems, for targets specifically in oncology, alongside higher molecular weight complexes and integral membrane proteins for structural and biophysical analysis. Demonstrable track record and deep knowledge of leading a group that routinely produces large scale protein preparations with targets purified to homogeneity across numerous FPLC systems - while leveraging multiple expression sources including bacterial, insect cell and mammalian suspension cell culture – with resultant samples subsequently being deployed across an array of discovery applications. Ability to direct and manage a large group of scientists efficiently and effectively. Excellent management and interpersonal skills.
