There is precedent for this. Check the reference below. This protein was 
crystalized from solution at a concentration of 150 mg/ml. Keep 
concentrating, I guess !

/In situ/ data collection is a good approach to try in your case. You 
could use the CrystalDirect technology for automated crystal harvesting 
that is more gentle to crystals than manual harvesting. This is  
available both at EMBL Grenoble and EMBL Hamburg facilities, which offer 
integrated crystallography services in collaboration with the ESRF and 
Petra III synchrotrons.

dissertation research. I have purified a complex of two proteins, and 
I can consistently grow crystals in 10% PEG3350, 0.2M KSCN, 0.1M 
BIS-TRIS propane pH 7.5. These crystals have sharp edges and can grow 
to a large size (greater than 0.5 mm), but the crystals seem to be 
very fragile. When we open the drops to harvest the crystals, we have 
little time to harvest the crystals before they crack. When we move 
the crystals to a cryoprotectant, over time they start fracturing. 
We've tried using different percentages of glycerol, ethylene glycol, 
PEG400, and oil for cryoprotectants with no success. Needless to say, 
the crystals do not diffract well, with spot patterns that look very 
streaky/mosaic, which I presume is due to the defects that we see in 
harvesting/handling. We have screened for alternate crystallization 
conditions, but we seem to get the same morphology in other 
conditions. Does anyone have suggestions for additives we could use 
post-crystallization to help stabilize our crystals?

Under the umbrella of the EMBL-ESRF Joint Structural Biology Group a 
collaborative project to upgrade MASSIF-1 and exploit synergies with the 
EMBL High Throughput Crystallization facility (HTX Lab) in Grenoble has 
been established. An integral part was the installation of one of the 
CrystalDirect automatic crystal harvesting robots into the beam line 
environment, enabling new experimental modalities. Thanks to the hard 
work of the EMBL Instrumentation, Synchrotron Crystallography and HTX 
Lab Teams, and the ESRF Structural Biology group, the installation is 
now completed.


We are now opening access to newly developed CrystalDirect plate-to-beam 
experimental modalities at MASSIF-1 via the EMBL HTX Lab. We are 
inviting applications for either collaborative or user access. 
Opportunities for funded access are available through iNEXT Discovery. 
If you are interested in any of the new experiments listed below please

•    Automated Protein-to-structure pipelines with CrystalDirect 
plate-to-beam cryogenic data collection. Either for full diffraction 
data collection or rapid diffraction-based screening and prioritization

identify conformational variability with potential insights into protein 
dynamics.


•    Automated crystal dehydration experiments to modulate crystal 
parameters.


Pure samples can be shipped to the EMBL HTX Lab were crystallization 
experiments will be carried out. Online experimental design and 
real-time access to results is facilitated via the Crystallographic 
Information Management System (CRIMS) web interface. Typical 
experimental workflows include crystallization screening and 
optimization, crystal harvesting and CrystalDirect-plate-to-beam data 
collection at MASSIF-1.

EMBL Grenoble is looking for a Data Scientist to develop innovative 
approaches for the analysis of structure guided drug design data based 
on advanced computing and machine learning technologies. The HTX lab at 
Grenoble provides access to fully automated crystallographic pipelines 
based on the CrystalDirect technology. These pipelines are applied to 
support large scale ligand and fragment screening projects and generate 
tens of terabytes of data yearly. Data and metadata is automatically

Postdoctoral opportunities: Structural analysis of alloantibody-HLA 
complexes by Cryo-EM.


This is part of a multidisciplinary, collaborative project between the 
Marquez (EMBL Grenoble), Leach (EMBL EBI) and Kosmoliaptsis (Cambridge 
University) laboratories.  The project aim is to understand molecular 
and structural basis of specific alloantibody-HLA interactions and their 
impact on organ transplant rejection. We employ biochemical, 
biophysical, structural and computational techniques to study specific 
alloantibody-HLA interactions that mediate different effector functions. 
This research project is directly motivated from real clinical problems 
and benefits from direct access to a clinical context (Kosmoliaptsis lab)


We are looking for intrinsically motivated, hypothesis-driven 
individuals with a PhD in relevant fields and significant experience in 
structural biology. Experience in single particle analysis by Cryo-EM 
will be he highly appreciated. The candidate will be based in Grenoble 
and travel to the UK for short time periods.

The ESRF-EMBL Joint Structural Biology Group (JSBG) invite COVID-19 
related proposals that can benefit from the seamless integration of the 
EMBL High throughput Crystallization Facility (HTX Lab) with the highly 
automated JSBG MX beamlines at the ESRF. The pipeline starts with 
mail-in samples and integrates crystallization screening, crystal 
optimization, ligand soaking if required and eventually fragment 
screening with libraries of up to 1200 fragments. Crystals obtained will 
be automatically harvested and passed to the ESRF for automated X-ray 
data collection, with real-time access to experimental results and 
parameters. This pipeline is uniquely suited to support structural 
biology projects in conditions of confinement and restricted access to 
labs. The HTX lab is already accepting crystallization projects on 
COVID-19 related research and three MX beamlines (MASSIF-1 (ID30A-1), 
ID30B and ID23-2) will become operational from mid-May.


Users wishing to benefit from this combined access for COVID-19

interest, whereupon a short application form will be sent for completion.

The High throughput Crystallization Facility at EMBL Grenoble (HTX Lab) 
provides access to a fully automated protein-to-structure pipeline based 
on the CrystalDirect technology and the CRIMS software. This pipeline 
starts with mail-in samples and integrates crystallization screening, 
crystal optimization and automated harvesting in a continuous and fully 
automated workflow that can be operated by any scientist from a desktop 
computer with connection to the internet. The CRIMS software is able to 
communicate with the ESRF and Petra III synchrotrons to support 
automated and remote X-ray data collection and provides real-time access 
to experimental results and parameters. These capabilities are uniquely 
suited to support structural biology projects in conditions of 
confinement. The HTX lab is accepting projects on COVID-19 related 
research starting April 16th.


The HTX Lab also provides access to dedicated pipelines for the rapid 
analysis of protein-small molecule interactions as well as for large 
scale fragment screening to support development of inhibitors and 
chemical tools. This includes access to two fragment libraries (1200 and 
700 fragments respectively) and automated data processing for hit 
identification.

The Marquez (EMBL-Grenoble), Kleywegt and Velankar (EMBL-EBI) groups
have a potential opening for a post-doctoral fellow in structural
bioinformatics through the EIPOD Fellowship program. If you're
interested, please contact José Marquez (marq...@embl.fr) before
September 7th (including a CV).

The successful candidate will be working in an interdisciplinary project
at both sites involving integration and archiving of high-throughput
crystallography data. Applicants should hold a PhD in physics,
mathematics, (bio-)chemistry, biology or similar, involving the
application of computational methods in the life sciences, and should
have experience with professional scientific software development
(design, implementation, and maintenance).

Requirements:
- fluency in Python and experience developing web-based applications
using the Python Django framework
- experience with object-oriented PHP
- experience with relational databases and SQL
- experience with JavaScript frameworks (such as jQuery)

Experience or familiarity with one or more of the following would also
be advantageous:
- a background in, or a good level of understanding of X-ray crystallography
- one or more higher-level programming languages (e.g., C++, Java, Fortran)

Automated pipelines for compound and fragment screening in Grenoble

The EMBL Grenoble, the ESRF and the iNEXT consortium are jointly
offering access to automated pipelines for compound and fragment
screening based on the CrystalDirect* technology and the MASSIF**
beamlines. Access to these pipelines can be obtained through the E.C.
H2020 project iNEXT (www.inext-eu.org). Applications will be reviewed by
the iNEXT scientific review panel in collaboration with the ESRF MX
Beamtime Allocation Panel.

These pipelines are offered in two forms:

Compound Screening Pipeline: Automated screening via X-ray
crystallography of a collection of small molecule ligands identified
through previous assays (i.e in silico, screening, activity assays, SPR,
etc.). Applicants must be able to provide the ligand collection, pure
protein in sufficient amounts and well-established and robust
crystallization conditions.

Fragment Screening pipeline: Automated large scale fragment screening
via X-ray crystallography. The fragment collection will normally be
provided by the Grenoble facility although screening of user provided
libraries is also possible. Applicants must be able to provide pure
protein in sufficient amounts and well-established and robust
crystallization conditions

Additionally, we are also offering access to an Integrated
Macromolecular Crystallography pipeline offering fully automated, remote
controlled access to crystallization screening, optimization of crystal
conditions, automated crystal mounting and cryocooling (based on EMBL'S
CrystalDirect technology and automated (or manual) data collection at
the ESRF MASSIF-1 or MASSIF-3 beamlines.

Access to crystallization services at the High Throughput 
Crystallization Laboratory of the EMBL Grenoble Outstation is supported 
by the E.C. Funded Biostruct-X project. To apply please go to the 
Biostruct-X http://www.biostruct-x.eu/ web page. Selecting Single 
application will open a pane giving access to PP  HTX.

Starting in April 2013 and in collaboration with ESRF BM14  beamline the 
services will be extended to include automated evaluation of crystal 
hits through in plate X-ray experiments. For this purpose,* 
*crystallization experiments will be performed in specially developed 
low-background CrystalDirect plates (/Acta Cryst./ 2012. D*68*, 
1393-1399). A crystal pointing tool in CRIMS allows user to select 
through the web the crystals you want to test.



The High Throughput Crystallization ( HTX ) laboratory of the EMBL 
Grenoble outstation is one of the key platforms of the Partnership for 
Structural Biology (PSB) http://www.psb-grenoble.eu/and is located 
next to the European Synchrotron Radiation Facility (ESRF) and the 
Institute Laue Langevin (ILL). This facility offers completely automated 
crystallization screening in hanging and sitting drop set up at two 
temperatures (20°C and 5°C). Results together with all experimental 
parameters are made available to users in real time through the 
CRystallization Information Management System (CRIMS), which includes 
fast and convenient web user interfaces providing advanced tools like 
drop scoring, drop history, navigation through project targets and 
samples and project summaries and a crystal pointing tool to support 
automated in plate x-ray diffraction experiments among others. 
Crystallization plates are made available to users upon requests 
(typically during their next synchrotron visit) and facilities for 
crystal mounting and freezing are provided. Since the start of 
operations the HTX lab in Grenoble has provided crystallization 
screening to more than 650 scientists.

*Job description:* The High Throughput Crystallization laboratory at the 
EMBL Grenoble Outstation is offering a posdoctoral position. The HTX Lab 
is actively involved in the development of new methods and concepts in 
macromolecular crystallography and works in close co-ordination with the 
high-throughput protein expression and synchrotron instrumentation 
groups at the outstation. The selected candidate will participate in the 
Eurostars funded Crystal Clear project, whose aim is to further develop 
a novel and promising approach to protein crystallization in 
collaboration with an industrial partner. He/she will work on 
crystallisation using the new method and determining crystal diffraction 
quality and will participate in the research projects of the laboratory. 
He/She will also coordinate activities with other Eurostar participants


*Qualifications and experience:* The ideal candidate will have a PhD in 
structural biology or a related subject. The candidate should have 
significant experience in protein crystallization as well as good 
knowledge of X-ray crystallography methods. He/she should have good 
communication skills; strong motivation and excellent scientific skills 
are essential, as well as willingness to work effectively alone and in a 
team. Desire to work in an multicultural  interdisciplinary environment 
is essential.


*Contract:* An initial contract of 1 year will be offered to the 
successful candidate. This can be renewed, depending on circumstances at 
the time of review.
