Hi Colleen,
I was involved in an Immunohistochemical study of the nerves in the human
ureter that used Zamboni's fixative and cryostat sections. This study was
before the invention of HIER.
If you are using Zamboni-fixed, paraffin embedded tissue then you will probably
need to do HIER before
Hello Histonet,
Can you give me feedback on trying to do IHC on samples fixed in Zamboni's
fixative? I have an idea but need confirmation .
Thank you in advance

Laboratories confirm assay performance when conditions change that may affect
performance.
NOTE: A change in antibody clone requires full revalidation/verification of the

Good afternoon,
I would like to inquire what everyone's decal (RBD and/or Decal I) changing
routines are. We are trying to determine if we should continue to change
weekly, or based on how many specimens we put in it, or what the pH is. Any
help is greatly appreciated, thanks!
Jennifer Wooten,
Hi Histopeeps!
I hope you are having a great week. I wanted to drop a quick post because I
have some really exciting opportunities to share.
Why are they exciting?
1. Beautiful locations
2. Day shift hours
3. Excellent compensation packages
4. They are ready to interview and
Good morning. I would like to obtain positive control blocks for
Helicobacter pylori. Thanks in advance for your help.

I’m not completely sure but I think you can do either a mini validation or a
comparison validation to show there’s no difference between the results.
I think CAP has a requirement for what to do when reagents change.

Hello,
Kindly remove me for the Histonet mailing list. I've attempted several
times with no success.

Scott:I’m pretty certain that you wouldn’t need to completely re-validate after
changing diluents, but you may have to change the dilution factor slightly.
It’s my understanding that a change like this only requires staining a few
known positive samples, verifying that the results you obtain
Hi Scott,
It may be best to call CAP to get an interpretation. However, I think that
since you would be changing diluent for predictive markers, a full validation
may be required.

What is everyone's take on what needs to be done from a verification/validation
standpoint if you switch diluents for your concentrated antibodies? The CAP
summary of recommendations on the validation IHC antibodies doesn't
specifically mention the changing of diluent as one of the variables.
