Will not work with NAT between the two sites (unless it is a one to one
NAT without firewalling).

Still I m bit confused.
If I configure dial plan than it means My sipx site 2 should be on the
static ip address.
Will it work fine if Nat is in b/w the 2 Sites.
Should i Have to specify 5060 port in gatway registration.

It depends on whether you want the dialing to be "native" or "prefixed".
This is based ona  3 digit dialplan.

Site 1 - Extensions 200-299
Site 2 - Extensions 300-399

Create a gateway with the IP of site 2's IP address.
Create a rule at site 1, CUSTOM, prefix 3 and 2 digits. Resulting call
entire dialed number, use gateway just created.

Do the same thing at site 2, prefix 2 and 3 digits. Similarly you can
create a rule to prefix "8" so you can transfer to VM on the other
system, etc.

IF both sites are in the same range (200-299) then it's pretty simple to
make the user dial a prefix like "4" in the dial rule and send just the
3 digits (dialed number prefix is 7 and 3 digits, resulting call is and
append matched suffix, etc., so 7200 transfers to voice mail on the
opposite system).
