Richard, I don't know if has any bearing on your problem with TM but I had a similar problem this morning.
I'd run out of space on my storage drive and moved some files (195GB) to free up space. When TM went to do the next back-up it failed. I realised that I'd not 'Excluded' that particular folder in TM Prefs and it now backs-up fine. Don't know if it will help but... I took the drive out of Spotlight's list but that did not seem to help. I also used Disk Utility to Repair the disk. It took ages but did not report any problems at the end. I then used Drive Genius to defrag (what is going on with that huge spinning disk graphic?). Unfortunately this seems to have put Time Machine completely out of action. It is asking me to set it up again. The partition it is on will not mount or be repaired ("First Aid failed") in Disk Utility. It looks like I will have to erase it and start again!
