When running the simulation you can activate rerouting. To add randomness you can add a random factor to the edge weights. See http://sumo.sourceforge.net/userdoc/Demand/Automatic_Routing.html and in particular http://sumo.sourceforge.net/userdoc/Demand/Automatic_Routing.html#Randomness
