Which file is huge? Is it in your Mail folder...try emptying Junk and
Trash then compress folders Inbox, Sent, Trash...then check again.

My guess is the Sent folder. If you routinely email attachments, then all those messages (with the attachments) will be in your Sent folder, unless you move the messages to other folders. Even then, the attachments will still be there.

In my experience, most of the time, there's no real point in keeping copies of attached files in your mail store, because you already have the originals. This particular situation is where I think that Mozilla's email clients (both Seamonkey and Thunderbird) really shine, in that there is the capacity of deleting attachments from a message.

When viewing a message, if you right-click on an attachment, there are options for Detach, and Delete (as well as the more familiar "Save as"). If you Delete, then the attachment will be deleted from your mail store. If you select Detach, it's essentially a combination of "Save as" and Delete, where the attachment is saved to a user-specified location, and then deleted from the message. With a detach, as long as the saved file isn't relocated, then it's still associated with the message.

My suspicion is that these features were built to support IMAP connections, in the days when IMAP user quotas were typically a lot smaller than they are now. If you're in that kind of situation, getting attachments out of your mail store really makes a difference in how much space you have.

However, even with POP setup, I consider it useful to delete attachments from sent messages, because keeping the attachments merely puts duplicate copies of those attachments in my mail store, and there's no compelling reason for that.

Clean out your sent messages by killing off the attachments, and you'll likely have a noticeably smaller profile.

If you're in the process of moving your mail store to a different drive, that's probably reason not to do much in the way of detaching (unless you're confident that your detached files won't be relocated), although you may find that you have some messages, where you want the cover message, but where you don't really need the attachment, and can afford to delete.
