By signature you mean the following lines, right?

If you hardcode the word "Zipper" and its email address in the reply
template, then no, TB! won't change it. If you use the macro
%FromFName and %FromAddr, and you change the Account before typing
anything in the message body, then they'll be changed.
In other words, changing Account from [Options][Active Accoutn] will
not change the template for the message, which is decided when you
"create" the message, but it'll change the macros used in the
template.
