With a server side version of TiddlyWiki such as TiddlyWeb the might
be more achievable. Ben Gillies and I wrote some server side code that
pulled from an smtp email account into a TiddlyWeb bag.

However as Christian says sooner or later you will hit 300mb files.

My beefcake plugin (http://svn.tiddlywiki.org/Trunk/contributors/
JonRobson/plugins/beefcake/beefcake.tid) would help ease the loading
times by allowing you to suck in emails as you need them, but it's not
tried and tested on really really big files.

Put simply if you wanted to go down this route, you probably could but
you would encounter lots of unsolved TiddlyWiki problems and would be
pushing the tools boundaries. If you like a challenge maybe you still
want to but I wouldn't recommend it :)
