Yes, you should do

/etc/init.d/cups stop; nano /etc/cups/printers.conf;
/etc/init.d/cups/start

CUPS is reading /etc/cups/printers.conf shortly after it starts. While
running it writes the printers.conf which it has read back, with
changes, for example if a printer is enabled or disabled. So if you are
editing printers.conf while CUPS is running and without immediately
restarting CUPS after editing, your changes will not be seen by CUPS and
will get overwritten next time CUPS writes printers.conf. Therfore
always stop CUPS before editing printers.conf and start it again
afterwards.

Can you try it this way and tell whether it solves your problem? Thanks.
